<i>Plasmodium</i>-infected mosquito bite elicits an immune response at the bite site

Abstract To induce long-lasting immunity, the host’s immune system must recognize and respond to an invading pathogen upon first encounter. Plasmodium sporozoites, infective stage of malaria parasite, are inoculated into skin by infected mosquitoes as they probe for blood. Only 20% sporozoites reach liver, while rest either draining lymph nodes (dLN) or remain in area at least 6 hours, shedding abundant amount their major surface protein, circumsporozoite protein (CSP). Yet, sterile immunity against CSP does not occur, despite presence foreign antigens dLN, repeated exposures, suggesting that may modulate host response. In this study we investigate response elicited P. berghei-infected mosquito bite site in-vivo. Using intravital imaging inoculation a LysM-CRE GFP expressing mouse, can observe neutrophil recruitment real time. When investigating expression pro- anti-inflammatory genes skin, find specific patten is elevated bite, when compared uninfected mosquito.
